Parthenogenesis and activation of mammalian oocytes for in vitro embryo production: A review
نویسندگان
چکیده
Parthenogenesis is a form of asexual reproduction found in females, where growth and development of embryos occurs without fertilization by a male. Parthenogenesis occurs naturally in aphids, Daphnia, rotifers, nematodes and some other invertebrates but can also be induced efficiently in mammalian oocytes by providing appropriate stimuli in-vitro. Recently, parthenogenesis has attracted wide attention because of the role of activated oocytes in the field of research that have been described such as intra cytoplasmic sperm injection, cloning by nuclear transfer, somatic cell cloning, investigating culture conditions etc. & potential for deriving pluripotent stem cell lines and their differentiation into various cell lines that can be utilized for various tissue engineering applications. The parthenogenetically activated oocytes possess maternal genome and can developed in to either haploid, diploid or polyploidy embryos with the help of it we can analyze the possible role of all the genes involved in imprinting processes as well as the role the paternal genome plays during early embryo development by comparing them with fertilized embryos. Several methods are able to induce parthenogenetic activation through the elevation of cytoplasmic free calcium in oocytes. But one common, universal method or activation agents has not been developed for all species because the process is highly specific for each species. Therefore, activation step for each species need to be optimized accordingly. This review describes the general method of activation of mammalian oocytes and their genomic imprinting ana-

P-70: The Effect of Hydrostatic Pressure on Parthenogenetic Activation of Mouse Oocytes Derived from In vitro Grown Ovarian Follicles
Background: Parthenogenesis is the production of an embryo from a female gamete in the absence of any contribution from a male gamete, with or without the eventual development into an adult. In vivo, mammalian parthenogenesis is a rare event. Parthenogenesis can be efficiently induced in vitro with a variety of mechanical, chemical, and electrical stimuli in several species. Hydrostatic pressur...
متن کاملP-96: Mechanical Activation of Parthenogenesis in Mouse Oocytes Using Hydrostatic Pressure
Effective protocols are introduced for parthenogenesis activation in oocytes. Hydrostatic pressure can act as a mechanical stimulator that rearranges egg contents, leading to new structural or molecular combination. Alternatively, mechanical stimulation could stimulate a mechanically-gated process, such as opening or closing of stretch activated ion channels. This study, investigated the use of...

The present study was undertaken to assess the effect of different chemical activators along with 6-DMAP on in vitro matured caprine oocytes. From 4332 ovaries, 14235 cumulus oocyte complexes (COCs) were collected which were matured in TCM-199 medium containing follicle stimulating hormone (FSH) (5 µg/ml), Leutinizing hormone (LH) (10 µg/ml), oestradiol-17β (1 µg/ml) supplemented with 10% fetal...

Purpose: Parthenogenetic activation is among the crucial steps determining successful development of mammalian cloned embryos. This study, therefore, was conducted to evaluate the efficiency of a novel combined electrical-chemical artificial activation method for bovine cloning. Materials and Methods: In vitro matured bovine oocytes than were initially exposed to electrical pulse, used for cel...
متن کاملReactive oxygen species generation and use of antioxidants during in vitro maturation of oocytes: a Review
In vitro maturation (IVM) is emerging as a popular technology at the forefront of fertility treatment and preservation. However, standard in vitro culture conditions usually increase reactive oxygen species (ROS), which have been implicated as one of the major causes for reduced embryonic developmental competence. It is well-known that higher than physiological levels of ROS triggers granulosa ...
